Dostarlimab Vanishes Cancer Miraculously In Patients For First ... (Outlook India)

Dostarlimab, a drug was given in a small clinical trial to 18 patients for around six months, and in the end, it was found that cancer had just vanished ...

They were all in similar stages of their cancer - it was locally advanced in the rectum but had not spread to other organs. Speaking to the media outlet, Dr Alan P. Venook, who is a colorectal cancer specialist at the University of California, said that the complete remission in every single patient is “unheard-of”. “There were a lot of happy tears,” she told the New York Times. According to New York Times report, in a small clinical trial, 18 patients were given a drug named Dostarlimab for around six months. Five centuries later, an Indian author won a major literary award in the UK and shared it with her American translator, making it an apt occasion to discuss the status of translation in world literature. For a majority of publishing houses, both indie and big, translation work has to be subsidised.

Cancer disappears in patients 'first time in history' after drug trial ... (India TV)

Reportedly, cancer completely disappeared in the patients. It was undetectable by physical exam; endoscopy; positron emission tomography or PET scans or MRI ...

“There were a lot of happy tears,” she told the New York Times. Dostarlimab is a drug with laboratory-produced molecules that function as substitute antibodies in the human body. They were all in similar stages of their cancer - it was locally advanced in the rectum but had not spread to other organs.

'Unprecedented' 100% of First 14 Untreated Patients With Rectal ... (The ASCO Post)

In a study of patients with locally advanced mismatch repair–deficient (dMMR) rectal cancer, 6 months of treatment with the anti–PD-1 agent dostarlimab-gxly ...

Clinical complete response was indicated by an endoscopic visual exam showing disappearance of the primary tumor along with a normal digital rectal exam; the formal criteria for rectal MRI response also had to be met. “I’d like to highlight that the majority of these patients had big bulky tumors, and 94% were node-positive. Patients received dostarlimab at 500 mg intravenously every 3 weeks for 6 months and then underwent radiologic and endoscopic evaluation. So far, dostarlimab has spared every one of these patients of the need for chemotherapy, radiation, or surgery. The single-arm phase II study will ultimately enroll 30 patients with clinical stage II and III dMMR rectal cancer. Of the 18 patients enrolled to date, most (78%) have T3 or T4 rectal tumors.

What Is Dostarlimab, the Drug Used in 'Unprecedented' Cancer Trial? (NBC New York)

A GlaxoSmithKline monoclonal antibody called dostarlimab, or Jemperli, achieved an "unheard-of" result in a New York City cancer trial.

The term became more widely known in the last two years as a variety of monoclonal antibodies came out to treat COVID-19. First approved in the United States for use as a cancer treatment in early 2021, dostarlimab is a monoclonal antibody. Developed by a biotech company out of Massachusetts called Tesaro before being acquired by GlaxoSmithKline in 2019, dostarlimab is also known by the brand name Jemparli.

Unfortunately, for many patients this gamut of treatments comes with long-lasting consequences that can last the rest of their life. Ultimately, the trial is expected to include about 30 patients. "And since they couldn't find any signs of cancer, Dr. Cercek said there was no reason to make me endure radiation therapy." "Whether the results of this small study conducted at Memorial Sloan Kettering Cancer Center will be generalizable to a broader population of patients with rectal cancer is also not known." "We thought, 'Let's try it before cancer metastasizes as a first line of treatment'." "Very little is known about the duration of time needed to find out whether a clinical complete response to dostarlimab equates to cure," Sanoff explains, noting that we also need larger-scale replication of the results to be sure of the drug's benefits, which so far have only been seen in a minority of patients with MMRd tumors.

Dostarlimab Elicits Clinical Complete Response Among All Treated ... (OncLive)

Single-agent dostarlimab-gxly elicited a clinical complete response rate of 100% with no evidence of residual tumor among 14 patients with stage II/III ...

“It is going to be critical that we identify predictive biomarkers of pathologic complete response to help guide the treatment for our patients.” A cCR was defined as no evidence of residual tumor on both the digital and endoscopic rectal exams and on the rectal MRI results. The second patient was a woman aged 30 years who presented with approximately 3 months of rectal bleeding, a change in the caliber of her stool, and pelvic pain for approximately 2 weeks prior to diagnosis. Her final examination, after completion of 6 months of therapy was notable for a cCR and I’m very happy to say that this has been sustained now in follow-up for nearly 2 years.” Cercek presented an overview of the first 2 patients enrolled and treated with dostarlimab. A majority were women (67%) and had a median age of 54 years (range, 26-78). In terms of tumor staging 22% had T1/2 tumors and 78% had T3 or T4 tumors. The first patient was a woman aged 38 years who presented with a large, node-positive tumor. [We have] quite a bit of a range from 0.7 to nearly 24 months and all patients remain disease free,” Cercek said. At baseline and through treatment, patients were closely monitored and assessed for response at 6 weeks, 3 months, and 6 months. “No patients have required chemotherapy, radiation, or surgery [and] there has been no disease recurrence observed during the follow-up period,” Andrea Cercek, MD, said in a presentation of the data. Specifically, there must be visual evidence of primary rectal tumor disappearance on a normal digital exam and lack of signal on diffusion-weighted imaging with scar on T2-weighted imaging. “Longer follow-up is certainly required to establish the durability of this treatment.
